I have found that the best way (for me anyway) is to back up to a portable external hard drive as I have a laptop and not a desktop.I have an Iomega eGo 160Gb one which does me fine and I just use Vista`s backup facility rather than using the software that comes with it . Usually takes about 45mins but I keep a tight reign on what I install so as to keep the number of files to a minimum . Obviously backup times depend on how much you`ve got on your PC .I think if I was a movie and music buff I would need a larger one.
I also copy and paste individual files to a CD-RW disc.

I use a flash drive for currant stuff like my Resume, or Assignments, and then a combination of External HD and DVDs for longer term. I don't use any kind of automated system, I just burn the contents of my documents folder to a disk or external hard drive, then move those files to a folder named "Backed Up".
